要旨: Steady state cell turnover in the lung is very low. However, the organ has an impressive capacity for repair after injuries caused by exposure to toxic agents or viral infection. My lab has used a variety of techniques‒ in vivo lineage tracing, conditional gene manipulation, 3D organoid culture, and small molecule screens ‒ to identify genes and pathways regulating the regeneration of the ciliated and secretory luminal cells of the upper airways of the mouse lung from a population of unpolarized basal stem cells. We have evidence that the differentiation of the basal cells, and changes in their shape, adhesion and polarity, are coordinated by a network of genes controlled by the grainyhead-like transcription factor, Grhl2. One challenge has been to translate our findings with mouse models to human airways. To overcome this problem we have used primary human airway basal cells cultured under conditions in which they generate a polarized mucociliary epithelium with barrier function or clonal organoids containing ciliated, secretory and basal cells. This has been combined with Cas9-nuclease gene editing technology to test the function of multiple genes predicted to regulate cell fate and behavior during the regenerative process.
