The department was founded in 1969 to contribute to human welfare through cancer treatment research.

History

This division, founded in 1969, is one of the oldest department of medical oncology in universities in Japan . The department's first professor, Tatsuo Saito set up the criteria to evaluate the effects of cancer therapeutic agents clinically. During the first decade, a large number of drugs were developed and the clinical effects of these drugs were studied in phase trials introduced from the U. S by him. The effects of combination therapies were also examined, but the efficacy were limited. In 1978, As Dr.Saito's successor, Akira Wakui extended the research field into basic science. Intensive studies were carried out on drug sensitivity, drug resistance and drug delivery. Based on these studies, new strategies were developed and were applied in practice with remarkable effects in some cases. In 1991, Ryunosuke Kanamaru became the department's professor and in 1993, the Institute was reorganized as the Institute of Development, Aging and Cancer that now includes our department. In 2000,accompanied by the closure of the Attached hospital the clinical part was moved to the Affiliated Hospital of medical school. In 2003, Chikashi Ishioka became current research is focused on translational research of molecular oncology and chemotherapy including molecular-target therapy.

Research Interest
  1. Optimization of administration of anti-cancer drugs based on molecular diagnosis.

    Recent advances in molecular diagnostics such as sequence-based mutation analysis and genome-wide expression analysis would enable the novel approach to predict cancer chemosensitivity or drug-resistance, and therefore, efficient application of chemotherapy to cancer patients. Using DNA microarray analysis, we are developing predictive expression profiles that are highly correlated with specific chemotherapeutic treatments by using both tumor cell lines and cancer tissues from patients.

  2. Development of molecular diagnostics for cancer-risk assessment, origin of tumors unknown primary sites, and prediction of prognosis.

    Studies on the evaluation of cancer-risk for individuals are important for prevention and early detection of cancers. We have performed collaborative studies to develop systems on genetic testing for both familial breast cancer and hereditary colorectal cancers. Tumors unknown primary sites are not rare and approximately 20 patients visit to our clinical section every year. Because of unknown primary sites, initial trials of chemotherapy were often unsuccessful. We are trying to develop a diagnostic system to predict specific expression profiles similar to primary tumors by multiclass analysis.

  3. Studies on development of molecular target therapy for cancer.

    Current molecular target therapeutics mainly focused on receptor tyrosine kinases including, Bcr-Abl, c-kit, EGFR and CD20. Although these are successfully applied in cancer clinic, it is necessary to develop novel molecular targets and drugs that enable to modulate protein functions to regulate most of common cancers. We have focused on proteins derived from tumor suppressor genes (p53, PTEN and BRCA1) and human mismatch repair genes (MLH1 and MSH2) and have studied on possible mechanisms that restore or modulate the normal or mutated protein function to accelerate efficient cell death in cancer cells.

  4. Clinical studies on cancer chemotherapy of gastrointestinal malignancies.

    During last four decades, we have performed multiple studies on cancer chemotherapy for patients with gastrointestinal malignancies, including esophageal, stomach and colorectal cancers. At present, we have focused on i) clinical phase II study on chemo-radio therapy of stage II or III eshophageal cancer as a institutional collaborative study, ii) clinical phase II of advanced gastric cancer for irrinotecan and docetaxel combination chemotherapy, iii) other combination chemotherapy regimens for colorectal cancers and other primary tumors.

  5. Qualitative cancer imaging by positron emission tomography (PET) and its application to the evaluation of efficacy of cancer treatment.

    During last 2 decades, we have used 18FDG PET for qualitative diagnosis of tumors (especially gastrointestinal malignancies) and quantitative evaluation of responses against anti-cancer drugs. The efforts are now applying to the assessment of responses in novel molecular-targeted anticancer drugs.

  6. Studies on molecular mechanism of cancers using model mouse systems and their application to cancer chemoprevention.

    Using APC gene knockout mice and a mouse genetic system, we have studied on genetic and biochemical mechanisms underlying colorectal carcinogenesis. During this process, we have found novel genes that negatively modulate the carcinogenesis. The gene product and its molecular pathway have been considered as a novel molecular target for molecular and chemoprevention of human colorectal cancers.
